Default tuning protocols—Full Tuning and Instrument Check—cannot be modified or deleted, but custom tuning protocols can.
To create a custom tuning protocol
1 In Tuning Manager, under Protocols, click Add. A New Tuning Protocol tab is displayed in the workspace.
2 Double-click the Name field to modify the protocol name.
3 (Optional—CyTOF Administrators only) On the protocol, under General Parameters, click Apply Template to use a template other than Default Bead Tuning Template.
4 Under General Parameters, enter:
• Injection Volume (μL): ≥500 μL is recommended
• Pre-wash Cycle
5 Select the subcalibrations you want to include.
NOTE We recommend using the default settings for each subcalibration unless instructed by Fluidigm Service or Support for troubleshooting purposes.
6 Click Save.
To modify a custom tuning protocol
1 In Tuning Manager, under Protocols, click the tuning protocol you want to modify.
NOTE If you want to preserve the original tuning protocol, click Duplicate, and then modify the copy.
2 After the protocol is displayed, modify the settings that you want to change.
3 Click Save.
